The 4 to 6 leaf-age plantlets produced by seeds of Huashuang 3 and P34-3 lines were treated in artificial climate chambers at 6°C and 8°C for 32 and 42 days, respectively, and cultured in a 25°C culture room. All treated and control seedlings were transplanted directly from the test tube into the shade. In order to treat more seedlings in the flask at low temperature, the three-leaf-age test-tube plants regenerated from ex vivo microspores of F1-89 plants (having chromosomes doubled, the same below) were placed in artificial climate chambers at 5-6°C. The medium was treated for 14 d (T1 treatment) and 22 d (T2 treatment), and then cultured in a culture room at about 26° C. for 18 and 10 days, respectively, until development to a 5-leaf age and control without cold treatment.
In order to treat more plantlets in the artificial climate chamber at low temperature, we made a 3-leaf-age (3rd-leaf flat) seedling to be treated at a low temperature for a certain period of time and then cultured in a culture chamber at about 26°C for a period of time to allow it to develop into energy transfer. After planting alive strong seedlings, the vernalization feature was lost. As to whether the re-incubation time of 26°C above 18d caused the loss of the characteristics of vernalization, further experiments are needed.
Artificial climate chamber at the same treatment temperature, the flowering period with the extension of treatment time advance, 42d treatment than 32d treatment 1 ~ 4d ahead of schedule. Within the same treatment period, the temperature has a greater impact on flowering than processing time. Treatment 42d, 6 °C than 8 °C treatment ahead of 3 ~ 5d. In this experiment, the artificial low-temperature treatment promoted the early flowering of rapeseed and kissed the conclusions of previous researchers. The plantlets of semi-winter rape were treated in an artificial climate chamber at about 8°C for 32 days. All plants passed the vernalization stage.
